She is the daughter of the yesteryear actress Dina Pathak, who was well-known for her mother and grandmother roles in the films. Supriya was brought up in a traditional Gujarati family. Supriya's sister Ratna Pathak is also one of the eminent actresses and is married to the ace Bollywood actor Naseeruddin Shah.

Supriya is one of the most influential actresses in the industry. Supriya Pathak was mainly a theatre actress and she debuted in Bollywood in the year 1981 with the film 'Kalyug'.
